Surface Plasmon Resonance (SPR) is an optical-based label-free technique that provides real-time, quantitative data on interactions between molecules as an alternative to classic endpoint interaction techniques such as ELISA. With this powerful and cutting-edge technology accurate, precise, and robust assays can be developed to assess affinities, kinetic properties, potency, and active binding concentrations. SPR assays have the advantage of having a high throughput, flexibility, and sensitivity and provide a broad application such as potency measurements or molecule characterization studies.

SPR monitors binding events in real time between biomolecules by detecting changes in the refractive index at the surface of a sensor chip. As an example, for an antibody kinetic measurement, the target antibody is captured by an anti-IgG antibody that has been immobilised on the surface of the sensor chip. Different antigen concentrations are then sequentially injected onto the surface of the chip. During the interaction between the molecules, the SPR system generates a real-time sensorgram comprising three key phases: association, equilibrium, and dissociation. By fitting this data to a mathematical model, association (Ka) and dissociation (Kd) rate constants can be calculated to determine binding affinity. As the setup can be highly customized by capturing the target molecule on the sensor chip surface instead, using direct immobilization instead of capturing approach or using several flow cells to analyze different interactions in parallel, the most optimal setup can be developed for your specific molecule.
